Description: We designed a helix to contact the ligand, but the helix needs some back up to stay in place. You can mutate many of the residues to create a more stable configuration and make a more active catalyst (ie create a Helix-Loop-Helix). This puzzle will be worth 125 global points! if you are new to Foldit, make sure you have completed the Introductory Design Puzzles (6-1 through 6-4)

we don't want you to be influenced by those solutions and want to see what you can come up with giving this starting puzzle (which is different that the starting puzzle from Back Me Up 1)

While it's possible that the best one will be a helix-loop-helix like the starting structure, we're also very interested in finding out if the Foldit community can come up with an even better geometry. So if you find something that gives you a better score, go for it.

An inquiry

A question about the internal area. It appears to be missing - a rather large void in there. Some team solutions will have a void in this area while others show no void throughout this vast space.

Team member (gdnskye) - "..i can see the shape of the backbone above seg 239 follows the iso shading, just it seems a very big gap, and i wasnt sure if the low starting score in seg 46 was something to do with it all."

can you explain a bit more what's going on in there?

probably not worth trying too hard...

Lysine 239 is probably moving around a lot because its surface exposed, so it's probably not worth pursuing unless you can find some good scores that way.

The reason we locked many of these exposed residues (instead of letting you shake them) is because otherwise doing a shake or wiggle sidechains would get very slow since this is a large protein.
