Description: | Activation of small GTPase Ras is a critical step in signal transduction pathways leading to the expression of genes involved in cell growth, differentiation, and survival. When functioning properly, GTPase Ras is a key regulator of cell function. A mutation of Ras genes, however, can lead to (over)production of invariably active Ras proteins, potentially leading to cancer as well as other diseases. Your job is to find the native structure of GTPase Ras to better understand its conformational abilities. Keep in mind that the GTP-bound conformation of this protein should be in an ON state, signaling cell growth. The most stable of designs will be used in a subsequent puzzle where the goal is then to bind to GTP most efficiently. Keeping this follow-up puzzle in mind, try out some mutations in your design as the literature shows that certain mutations have a much higher affinity for binding and staying bound to GTP.
