Description: | This is a follow-up puzzle for Puzzles 1369 and 1372. Here we've provided the top Rosetta prediction as the starting structure. Note that predicted contacts are still available, and can be accessed from the Main menu (Selection Interface) or in the Actions tab (Classic Interface). Players may load in manual saves from Puzzles 1369 and 1372 and use them as a starting point here.
This is a domain of the human dysferlin protein. Dysferlin is found in muscle cells and associates with the cell membrane, although its exact function is unknown. Mutations in the dysferlin gene can cause a debilitating type of muscular dystrophy. See the blog for more info. Our collaborators at the Jain Foundation would like to model the structure of dysferlin to better understand its function, and have asked Foldit players to help! Players may recall folding the C2B calcium-binding domain of dysferlin in Puzzles 1291 and 1293b. The residues modeled in this puzzle link two calcium-binding domains in dysferlin. It is unclear whether this domain serves a particular function, but some experimental data suggests it is well-folded. Sequence:
