Description: Like Puzzle 1048, this puzzle is aimed at designing short peptides that fold into stable structures, now with an emphasis on designed sheets! See the puzzle comments for more information about filters. The Baker lab will run folding predictions on your best designs, and the the ones that perform well will be synthesized and further validated for structural stability. Remember, you can use the Upload for Scientists button for up to 5 designs that you want us to look at, even if they are not the best-scoring solutions!

Residue IE Score: Monitors that all PHE, TYR, and TRP residues are scoring well.

Core Existence: Ensures that at least 10% of the residues are buried in the core of your design.

Fragment Score: Scans for parts of your design that are unlikely to fold up naturally. Use Rebuild and Remix tools to fix these regions of your design!

Disulfide Filter: Yields a bonus of up to 1000 points for 2 or 3 disulfide bonds. More than 3 disulfides will incur penalties.

Secondary Structure: Checks that at least 6 residues form sheets; penalties are incurred if fewer than 6 residues form sheets.
